Anionic regions in nuclear proteins

چکیده

Recent technological advances have generated a flood of protein sequence data, much of it deduced from the DNA sequences of cloned genes. In this commentary I will discuss an unexpected sequence motif found in a number of proteins that are thought to interact directly or indirectly with DNA. The motif, which I will term an Aregion (acidic region), is a local high concentration of acidic residues, which may in extreme cases take the form of monotonous runs of either glutamic or aspartic acid residues. I will list below a number of proteins that have been shown to contain Aregions, and speculate about the possible role of such regions in vivo.

Purification and reconstitution of the connexin43 carboxyl terminus attached to the 4th transmembrane domain in detergent micelles.

In recent years, reports have identified that many eukaryotic proteins contain disordered regions spanning greater than 30 consecutive residues in length. In particular, a number of these intrinsically disordered regions occur in the cytoplasmic segments of plasma membrane proteins. These intrinsically disordered regions play important roles in cell signaling events, as they are sites for prote...

Small, anionic, and charge-neutralizing propeptide fragments of zymogens are antimicrobial.

Some inactive precursor proteins, or zymogens, contain small, amino terminus, homopolymeric regions of Asp that neutralize the cationic charge of the active protein during synthesis. After posttranslational cleavage, the anionic propeptide fragment may exhibit antimicrobial activity. To demonstrate this, ovine trypsinogen activation peptide, and frog (Xenopus laevis) PYL activation peptide, bot...

Molecular Docking of Curcumin With Breast Cancer Cell Line Proteins

Background: Breast cancer is known as the most widely recognized dangerous tumors; therefore, the most common reason for mortality among all instances of harmful neoplastic illness in females. This is because the lack of specific signs and symptoms at the early stage and at the aggressive nature. Currently, breast cancer treatment such as chemotherapy, surgery and radiotherapy has not been effe...

Thermotropic lateral translational motion of intramembrane particles in the inner mitochondrial membrane and its inhibition by artificial peripheral proteins

Freeze fracturing and deep etching have been used to study thermotropic lateral translational motion of intramembrane particles and membrane surface anionic groups in the inner mitochondrial membrane. When the inner membrane is equilibrated at low temperature, the fracture faces of both halves of the membrane reveal a lateral separation between intramembrane particles and particle free, large s...
